Description of Services

DNA and RNA extractions, Realtime PCR analysis, SNP analysis using Sequenoms iPLEX system, expression analysis using the Taqman system. Manual extraction of RNA and DNA. We utilize our proprietary DNA extraction reagents, DNAQuick for DNA extraction from most of the sources. We also utilize Qiagen column protocols for some sample types. The Research Cell Bank (RCB) is a qualified, experienced laboratory that has been actively engaged in clinical research through B-lymphoblastoid cell (B-LCL) transformation and maintenance, DNA extraction, and inventory control for more than 30 years. The RCB is engaged in contractual, fee-for-service work for external entities requiring a large laboratory with expertise in the processing of blood and tissue samples, production of B-LCL, DNA extraction, cryopreservation, and maintenance of a large-scale frozen repository.

Applications:

Purification of DNA using Oragene is only done manually utilizing a modified salting out protocol with 5 Prime reagents. Isolated genomic DNA is rehydrated in T/E buffer at room temperature. The concentration of DNA is determined by a Spectramax 384 Plus spectrophotometer optical density (OD) measurement at 260 nm; the relative purity by OD 260/280 ratio.

Genetic Repositories Australia (GRA) is a national genetic repository for DNA and cell lines derived from appropriately consented disease-specific and population-based studies. GRA has been supported by an NHMRC Enabling Facility Grant and is based at the Prince of Wales Medical Research Institute (POWMRI).

GRA provides researchers with a central facility for the processing, long-term secure storage and distribution of human genetic samples (DNA and cell lines). This includes the production and provision of immortalised lymphoblast cell lines and DNA samples.

Description of Services

The Georgia Genomics Facility (GGF) is a sequencing and genotyping laboratory which provides services worldwide. The services we offer include capillary Sanger sequencing as well as high-throughput Roche 454 sequencing. We also offer various genotyping services: fragment analyses (microsatellite DNA and AFLP) are run on the ABI 3730xl, whereas we have various platforms for SNP genotyping (Illumina BeadXpress, TaqMan Open Array, and Real-Time PCR instrument). OGT was founded in 1995 by the pioneer of microarray technologies, Professor Sir Edwin Southern. The key focus areas of OGT are high throughput microarray and biomarker discovery. OGT also has an extensive range of cytogenetic products and services, including high resolution oligonucleotide arrays, labeling kits and our analysis software, Interpret.

Sample Management: management of collections of large numbers of biological samples for the use in scientific investigation. We can manage all aspects of sample collection from initial processing (e.g. DNA extraction, plasma/serum separation), long term storage (-80C, -20C) and distribution of resultant samples to third party clients if required. We have expertise in the collection and management of associated clinical data via our website and collaboration management tool. All our laboratory procedures are fully audit trailed using our LIMS system and we are ISO9001:2000 accredited.

The Facility provides sample preparation, assay design, lab work, and initial data analysis for a variety of SNP genotyping platforms including custom TaqMan, Beckman SNPstream, and Illumina GoldenGate, as well as whole-genome genotyping by Illumina Infinium and Affymetrix GeneChip systems. Diagnostic testing with DNA or RNA-based assays are developed and performed for clinical research applications.
