Study title: Neurocognition, Genetics and Adolescent Substance Abuse

Study abstract: Two major issues in substance abuse research are concerned with 1) neurocognitive mechanisms constituting vulnerability to substance abuse/dependence and 2) the impact of abused substances on the developing brain. In previous human research, these two aspects remained largely confounded since many studies used adult samples of drug abusers, which precluded the isolation of pre-existing vulnerabilities and consequences of chronic substance use. Genetic influences on substance abuse vulnerability are well documented, and recent research suggests a common genetic liability to abuse of different substances and a broad range of poorly controlled behaviors collectively labeled as "behavioral disinhibition". However, precise neural mechanisms and cognitive processes mediating genetic influences on behavioral disinhibition and, ultimately, on substance abuse remain poorly understood. We hypothesize that neural mechanisms of cognitive control play a critical role in the etiological pathways from genes to substance abuse vulnerability. To date, research on neurobiological and genetic aspects of behavioral regulation evolved independently, and few studies have linked genetics, brain function, and behavior in order to understand addiction etiology. This study seeks to reduce the existing gaps in knowledge through the implementation of a prospective longitudinal and genetically informative design with a model-driven choice of experimental paradigms that (i) tap into specific processes of cognitive control and (ii) have been linked to functionally defined neural substrates. Cognitive neuroscience research has identified electrophysiological and behavioral indices of cognitive control, including response inhibition, action monitoring, reward and punishment processing, executive attention, and decision-making and linked them to specific regions of the prefrontal and anterior cingulate cortex. During the initial funding period of this study, these measures were assessed in three consecutive cohorts of 12-years-old twins, i.e. prior to the typical onset of substance use (n=755), and longitudinal follow-up assessments are now in progress. This application seeks support for the extension of follow-up assessments till the age of 20, i.e. through the most vulnerable period of adolescence.

*Researchers with projects of all sizes are encouraged to submit as the $10,000 US in GenoFIND genomic services can be used for a small project or to cover part of the cost of a large project.

Eligible researchers must complete a simple award entry form and submit to DNA Genotek prior to 5:00 PM ET (GMT -5) on March 1, 2013.

Award Rules

  1. Applicant agrees that collected samples must be received for processing by May 10, 2013 and that applicant is responsible for all costs associated with shipping the samples to the laboratory, if selected as the winner.
  2. All submitted materials must be in English.
  3. Applicants who have a relative or member of their household currently employed by DNA Genotek are ineligible to enter.
  4. The GenoFIND Genomic Services award is open to researchers at academic institutions and non-profit research centers. All submitted samples and analysis must be for research purposes only.
  5. Applicant is responsible for ensuring that all necessary and legally required informed consents, authorizations, approvals, and permissions have been obtained in order to participate in this program, including any required Internal Review Board (IRB) approvals.
  6. Applicants must agree to DNA Genotek’s services terms and conditions.
  7. Applicants must allow DNA Genotek to publicly announce their name, project name or area of research, and institution or company if they are selected as the winner. Applicant must also provide feedback in the form of a testimonial or case study that may be used for marketing purposes (e.g. blog article).
  8. DNA Genotek will keep all information submitted in the award entry form confidential, except in the case of the winner.
  9. The award has no cash value and cannot be substituted for cash. The award is non transferable.
  10. DNA Genotek reserves the right to discontinue or modify the award at any time and for any reason. In such event, DNA Genotek will post a notice on this website. In the event of discontinuation of the award, DNA Genotek will have no liability to any existing or possible applicant or applicant’s employees or agents for such cancellation or modification.
  11. DNA Genotek may refuse to grant the award to any or all applicants if DNA Genotek determines at its sole discretion that the granting of such award would likely result in a violation of an applicable federal, state or local law, rule or policy.
  12. Each applicant is responsible for all of their own costs, and the costs of its employees and agents, that are incurred or that may be incurred in connection with this award, including, without limitation, any tax liability and any costs associated with entering into and participating in this award, developing ideas and materials, submitting materials, traveling and generally participating in the award.
  13. All decisions of the selection committee are final. The selection process is confidential.
  14. Applications will only be accepted for consideration if sent to DNA Genotek via the online entry form. All other applications will be rejected and destroyed.
  15. IN NO EVENT SHALL DNA GENOTEK, ITS OFFICERS, DIRECTORS, EMPLOYEES, SHAREHOLDERS, OR AGENTS, BE LIABLE FOR ANY DIRECT, INDIRECT, INCIDENTAL, SPECIAL, PUNITIVE, OR CONSEQUENTIAL DAMAGES WHATSOEVER ARISING UNDER OR AS A RESULT OF ENTRANCE IN OR PARTICIPATION IN THE AWARD PROGRAM, WHETHER BASED ON WARRANTY, CONTRACT, TORT, STRICT LIABILITY, OR ANY OTHER LEGAL THEORY, AND WHETHER OR NOT DNA GENOTEK IS ADVISED OF THE POSSIBILITY OF SUCH DAMAGES, THE FOREGOING LIMITATION OF LIABILITY SHALL APPLY TO THE FULLEST EXTENT PERMITTED BY LAW. THE TOTAL LIABILITY OF DNA GENOTEK ARISING UNDER OR RELATING TO THIS AWARD SHALL NOT EXCEED THE TOTAL VALUE OF THE PRIZE AWARDED TO THE SUCCESSFUL APPLICANT UNDER THIS AWARD.
